OBJETIVO: Avaliar os efeitos do treino físico imposto às ratas mães durante a gestação associado a uma dieta hipoproteica imposta durante os períodos gestacional e de lactação sobre o desenvolvimento e crescimento do fêmur da prole. MÉTODOS: Quarenta ratos machos Wistar, 90 dias de vida, foram divididos em quatro grupos: filhotes de mãe sedentária nutrida, filhotes de mãe sedentária desnutrida, filhotes de mãe treinada nutrida e filhotes de mãe treinada desnutrida, todos com n = 10. Treinamento físico em esteira durante 8 semanas, 5 semanas antes da concepção e 3 no período gestacional para as ratas mães dos grupos filhotes de mães treinada nutrida e treinada desnutrida. Indução da dieta hipoproteica às ratas mães durante a gestação e lactação para os grupos filhotes de mães sedentária desnutrida e treinada desnutrida. Após sacrifício, no 90º dia de vida, analisou-se peso, comprimento e conteúdo mineral ósseo do fêmur dos filhotes. RESULTADOS: Peso corporal, peso femoral e comprimento femoral diminuídos (p < 0,05) para os grupos filhotes de mães sedentária desnutrida e treinada desnutrida em relação aos grupos filhotes de mães sedentária nutrida e treinada nutrida, respectivamente. Não houve diferença no conteúdo mineral ósseo do fêmur em nenhum dos grupos. CONCLUSÃO: O treinamento físico leve em esteira durante a gestação não interfere no desenvolvimento e crescimento ósseo da prole. Entretanto, a desnutrição proteica durante esse período e durante a lactação promove prejuízos permanentes à estrutura óssea da prole.


OBJECTIVE: To evaluate the effects of physical training of mother rats during pregnancy associated with a low-protein diet offered during pregnancy and lactation on the development and growth of the femur of their offspring. METHODS: Forty 90-day old male Wistar rats were divided into four groups: pups of sedentary nourished mothers, pups of sedentary malnourished mothers, pups of trained nourished mothers, and pups of trained malnourished mothers; all groups included 10 rats. Physical training on a treadmill for 8 weeks, 5 weeks before conception and 3 weeks in the gestational period for mother rats of pups of trained nourished mothers and pups of trained malnourished mothers. Induction of low-protein diet to the mother rats during pregnancy and lactation for the groups of pups of sedentary malnourished mothers and trained malnourished mothers. After the pups were sacrificed, on the 90th day of life, we analyzed weight, length, and femoral bone mineral content. RESULTS: Decreased body weight, femur weight, and femur length (p < 0.05) were observed for the groups of pups of sedentary malnourished mothers and trained malnourished mothers in comparison with to the groups of pups of sedentary nourished mothers and trained nourished mothers, respectively. There was no difference in bone mineral content of the femur in either of the groups. CONCLUSION: Mild of physical training on the treadmill during pregnancy does not interfere with bone development and growth of the offspring. However, protein malnutrition during this period and during lactation promotes permanent damage to the bone structure of the offspring.

PURPOSE: To evaluate whether the neonatal leptin treatment during the first days of life can program the male reproductive organs weight and the lipid profile. METHODS: At birth 6 dams were divided into 2 groups: Leptin - each pup was injected with 50μL of recombinant rat leptin (80ng/g BW, sc), for the first 10 d of lactation; Control - each pup received the same volume of saline. After weaning, all pups received unlimited access to food until 190 days of age when they were killed. Values are given as mean ± SEM of 6 animals and Test t Student was used to analyze the results. RESULTS: The leptin treatment resulted in a significant increase in body weight (Control= 411.8±16.31; Leptin= 481.8±11.29, p=0.005) and food consumption (Control= 25.32±0.09; Leptin= 32.42±0.15, p=0.0001) and a significant reduction in triglycerides levels (Control= 540.0±117.9; Leptin= 93.25±15.21, p=0.006) and in the weight of hypothalamus (Control= 0.234±0.016; Leptin= 0.154±0.015, p=0.007), pituitary (Control= 0.104±0.0120; Leptin= 0.033±0.012, p=0.003), testis (Control= 3.75±0.055; Leptin= 3.19±0.10, p=0.002) and prostate (Control=1.641±0.1389; Leptin= 0.91±0.07, p=0.001). CONCLUSION: Leptin treatment on the first days of life can program the reproductive organs weight and the lipid profile of the progeny.


OBJETIVO: Avaliar se o tratamento neonatal com leptina durante os primeiros dias de vida poderia programar o peso dos orgãos do sistema reprodutor masculino e o perfil lipídico. MÉTODOS: Ao nascimento seis ratas-mãe foram distribuídas em dois grupos: Leptina - cada filhote recebeu 50μL de leptina recombinante (80ng/gPC, SC) nos primeiros 10 dias de lactação; Controle - cada filhote recebeu o mesmo volume de salina. Após o desmame, todos os filhotes tiveram acesso ilimitado a ração até 190 dias de idade quando foram mortos. Os dados são expressos como média ± erro padrão e foram analisados pelo teste t Student. RESULTADOS: O tratamento com leptina resultou em aumento significativo no peso corporal (Control= 411.8±16.31; Leptin= 481.8±11.29, p=0.005) e consumo alimentar (Control= 25.32±0.09; Leptin= 32.42±0.15, p=0.0001) e redução significativa nos níveis séricos de triglicerídeos (Control= 540.0±117.9; Leptin= 93.25±15.21, p=0.006), no peso do hipotálamo (Control= 0.234±0.016; Leptin= 0.154±0.015, p=0.007), hipófise (Control= 0.104±0.0120; Leptin= 0.033±0.012, p=0.003), testículo (Control= 3.75±0.055; Leptin= 3.19±0.10, p=0.002) e próstata (Control=1.641±0.1389; Leptin= 0.91±0.07, p=0.001). CONCLUSÃO: O tratamento com leptina nos primeiros dias de vida pode programar o peso dos órgãos do sistema reprodutor e o perfil lipídico da prole.

High levels of aggressive behaviors against intruders in the nest area are displayed by female rats during the first 10 days after delivery, declining thereafter to very low levels, even though lactation continues. Cross-fostering experiments were undertaken to test the hypothesis that pup age may affect aggression in lactating rats. The behavior of females on the 8th day after delivery when raising fostered 8-day-old pups was compared to that of females on the 8th postpartum day raising older pups (18 days old) for the last 5 days, and females on the 18th day after delivery raising fostered 18-day-old pups were compared to females in the same postpartum period nursing younger pups (8 days of age at the time of the maternal aggression test) for 5 days. Pup retrieval activity and plasma prolactin level were also analyzed. Females on the 8th postpartum day nursing 18-day-old pups were less aggressive than females in the same postpartum period, but with 8-day-old pups. Likewise, females on the 18th postpartum day nursing younger pups were more aggressive and presented higher levels of prolactin than females nursing older pups. Thus, pup development can alter the natural decline of maternal aggressive behavior

Cell proliferation and migration in the intestinal crypts, and cell migration in the villus are controlled by different mechanisms in adult rats. In the present study, weanling rats and fasting rats were used to quantitatively study the correlation of cell cycle parameters and epithelial cell migration in crypts and intestinal villi. Eighteen-day-old rats received a single injection of tritiated thymidine[3H]T dR (23:00 h); half of the pups were submitted to fasting 5 h earlier. Cell proliferation was determined in radioautographs of jejunal crypts, on the basis of the labeling indices (LI) taken 1,8,13 and 19 h after [3H]TdR. The results showed that the labeling index did not differ 1 h or 19 h after [3H]TdR between the fed (38.7 percent or 48 percent) and fasting groups (34.6 percent or 50.4 percent). The modified method of grain count halving indicated that cell cycle time did not differ between fed (16.5 h) and fasting rats (17.8 h); the growth fraction, however, had lower values in fasting (59 percent) than in fed rats (77 percent). Cell migration in the crypt, estimated by the LI obtained for each cell position, did not change with treatment. As for the villi, the cell migration rate was significantly retarded by 3 cell positions (8 percent). These results suggest that the cell migration in the villi of weanling pups does not depend directly on the cell proliferation and migration in the intestinal crypt, but is directly affected by the absence of food in the lumen.

Recent demonstrations of no changes in hypothalamic gonadotropin releasing hormone (GnRH) gene expression nd GnRH levels detected at the pituitary gland in diestrous and lactating rats, indicate that lactational hypogonadotropism in this species is not associated with inhibition of hypothalamic GnRH synthesis and secretion. Hypothalamic galanin potentiates GnRH effects on luteinizing hormone (LH) secretion in male and cycling rats. To explore the interaction between GnRH and galanin during lactation, we studied in vitro the effects of pulsatile stimulation with those peptides upon LH synthesis and secretion from rat pituitaries on diestrous 1 or day 10 of lactation. Hemipituitaries were separately incubated in 1 ml Dulbecco's Minimal Essential Medium supplemented with 1 per cent penicillin-streptomycin and fetal calf serum, at 37 degrees Celsius in 5 per cent CO2-air. The hemipituitaries were stimulated during 12 h with hourly pulses, 6 min each, of (a) gonadotropin releasing hormone (GnRH 25 ng/pulse), (b) rat galanin (600 ng/pulse), (c) GnRH plus galanin, or (d) saline. Medium was collected before each pulse to determine LH by radioimmunoassay. After the 12 h pulsatile regime total RNA was extracted and both actin and beta-LH mRNA were determined by reverse transcriptase polymerase chain reaction. There was a significant stimulation of LH secretion by GnRH (ANOVA, p<0.001) without significant differences between diestrous and lactation pituitaries. Galanin alone did not modify LH secretion but it potentiated the effect of GnRH upon pituitaries from diestrous (p=0.036) but not lactating rats. Neither peptide alone or its combination modified pituitary beta-LH mRNA levels. Results show that galanin regulates differently the secretion and synthesis of LH at the pituitary level. The disappearance of galanin-induced potentiation of GnRH effects upon LH secretion during lactation might contribute to the hypogonadotropism of lactation in the rat.

This study was designed to evaluate the thyroid and pituitary hormone levels in post-weaning rats whose dams were fed a low-protein diet during suckling (21 days). The dams and pups were divided into 2 groups:a control group fed a diet containing 22 per cent protein that supplies the necessary amount of protein for the rat and is the usual content of protein in most commercial rat chow, and a diet group fed a lowprotein (8 per cent) diet in which the protein was substituted by an isocaloric amount of starch. After weaning all dams and pups received the 22 per cent protein diet. Two hours before sacrifice of pups aged 21, 30 and 60 days, a tracer dose (0.6 mui) ofl25I was injected (ip) into each animal. Blood and thyroid glands of pups were collected for the determination of serum T4, T3 and TSH and radioiodine uptake. Low protein diet caused a slight decrease in radioiodine uptake at 21 days, and a significant decrease in T3 levels (l28 ñ 14 vs 74 ñ 9 ng/dl, P<0.05), while T4 levels did not change and TSH was increased slightly. At 30 days, T3 and TSH did not change while there was a significant increase in both T4 levels (4.8 ñ 0.3 vs 6.1 ñ 0.2 mug/dl, P<0.05) and in radioiodine uptake levels (0.34 ñ 0.02 vs 0.50 ñ 0.030 per cent/mg thyroid, P<0.05). At 60 days serum T3, T4 and TSH levels were normal, but radioiodine uptake was still significantly increased (0.33 ñ 0.02 vs 0.41 ñ 0.03 per cent/mg thyroid, P<0.05). Thus, it seems that protein malnutrition of the dams during suckling causes hypothyroidism in the pups at 21 days that has a compensatory mechanism increasing thyroid function after refeeding with a 22 per cent protein diet. The radioiodine uptake still remained altered at 60 days, when all the hormonal serum levels returned to the normal values, suggesting a permanent change in the thyroid function.
